I have never hunted the big 6 or 5, but think firearms regulations are different for different countries in Africa. Almost universal is a bullet diameter requirement of .375" or larger generating a minimum of roughly 3900 to 4000 fpe.
Costs can vary widely, depending on where the hunt is, even within the same country.
Some countries that should have all big 5 and maybe even big 6 would include:
Tanzania
Botswana
Mozambique (though not sure)
Zambia
Zimbabwe
Namibia's Caprivi strip bordering Botswana may have the animals, though I don't know if they all can be hunted legally.
South Africa may offer some of the Big 5 or 6 on ranch hunts.
HHK Safaris, Barrie Duckworth, John Sharp, Swainpoel & Scandroel, and others would offer those hunts.
Do a web search on any of those listed and look for prices. Those hunts can be extremely expensive, especially for lion and elephant. Daily rates going upto around 2,000 USD per day, 21 day minimum, and trophy fees approaching 10K or more for elephant. I don't know if anyone would book less than one month or even 45 days for all 6, though.
If you did a hunt for one or two plus plainsgame on a single safari, then back for another at a different time, etc... you may have better success and cheaper at any given time.
I have a friend that returned from an elephant hunt in Tazania that, I believe ran about 15K for two elephants, one bull and one tuskless. Pretty cheap considering the alternative hunts I've seen for elephant.
For hunts costing that kind of money, I'd definitely make contact with a good booking agent here in the U.S. and have them set it up for you. That would give you some recourse if things go wrong. There are a lot of booking agents on different African hunting forums on the web. Contact them and get a feel for what they offer and what you can expect. The agent won't cost you anything out of your pocket, they receive their commission from the outfitter or PH.

I just came back from a plains game hunt, and I can say "take money" because you are going to find more trophies that you want that you did not plan on taking in the first place. Choice of firearms is dictated by what you are hunting anf local legalities, check with your PH first. Since non-dangerous game does not have power limitations, I was lucky because arthritis has terminated much of my big bore shooting, however, a .30-06 with Nosler 220 grain slugs proved more than adequate for everything up to and including Kudu.
The best source of information I have found is Safari Club International (SCI), best fifty dollar investment I have ever made. Lots of information available via their websites, magazines, and meetings. If you can attend their shows you will meet PHs competing for your personal business, and they do compete. The Big 6/5 is expensive, and African hunting with an experienced PH is a real treat!! I am hooked. Definitely planning on going back.
